Hypothermisierende psychotrope Pharmaka als Strahlenschutzstoffe

Hypothermia producing psychotropic drugs as radioprotective agents

Abstract

Four different psychotropic drugs (Valium “Roche”, Taractan “Roche”, Tryptizol “MSD” and Insidon “Geigy”) have been tested as to their effect on O2-consumption and body temperature in mice, primarily from the viewpoint of their qualification as radioprotective agents. Within a certain range of doses, each substance depresses both O2-uptake and body temperature. A certain amount of protection against lethal irradiation with 800 r (survival rates were recorded for a period of 30 days) is exerted by the drugs if they are applied 15 minutes before initiation of the irradiation. It is not possible to decide whether this radioprotective effect is correlated more with inhibition of metabolism or more with lowering the body temperature. The striking relationship between inhibition of metabolism and radioprotection, which can be seen with taractan, seems to speak in favour of the leading role of metabolism. With exposure to whole-body irradiation 15 minutes after injection of this drug, the protective effect is slight; at the same time O2-consumption is lowered only moderately, although a reduction of body temperature occurs. Inhibition of metabolism reaches its maximum 1 1/2 hours after injection, whereby the extent of further body temperature reduction is comparatively small. If irradiation takes place at that time, the protective effect is highly significant. However, since hypothermia, depression of metabolism (hypometabolia) and hypoxia (general or local) are so intimately connected with each other and since they are all involved in the complex mechanism of the pharmacologically induced radioprotection in mammals, a decision such as mentioned above becomes rather superfluous.

Zusammenfassung

1. Vier neuere psychotrope Pharmaka,Valium, Taractan, Tryptizol undInsidon, wurden hinsichtlich ihrer Wirkung auf Gesamt-O2-Verbrauch und Körpertemperatur der Maus sowie ihrer Strahlenschutzwirkung gegenüber einer letalen Röntgenbestrahlung von 800 r untersucht.

2. Alle Stoffe senken in gewissen Dosen den O2-Verbrauch, und zwar Valium, Tryptizol und Insidon am stärksten in der ersten 1/4 Stunde nach der Injektion, Taractan erst 90 Minuten nach der Injektion.

3. Mit den Dosen der Pharmaka, die eine Senkung des O2-Verbrauchs herbeiführen, läßt sich im allgemeinen auch die Körpertemperatur vermindern, und zwar über die Dauer der stärksten Abnahme des O2-Verbrauchs hinaus. Insidon macht eine Ausnahme, indem hier die Körpertemperatur nur mit höchsten Dosen, die eher zu Steigerung des O2-Verbrauchs führen, herabgesetzt wird.

4. Alle Substanzen üben, wenn 1/4 Stunde nach der Injektion mit der Bestrahlung begonnen wird, einen Schutzeffekt (geprüft als %-Überleben in 30 Tagen) aus, der von etwa 10% bis 30% reicht und teilweise gut signifikant ist.

5. Wird bei Taractan die Bestrahlung zum Zeitpunkt der stärksten Stoffwechselhemmung (90 Minuten nach der Injektion) durchgeführt, verbessert sich mit den wirksamen Dosen der Schutzeffekt (%-Überleben in 30 Tagen) auf 40 bis 73% (mit außerordentlich guter statistischer Sicherung).

6. Auf Grund dieser Ergebnisse kann angenommen werden, daß bei einem pharmakologisch bewirkten Strahlenschutz Hypothermie und Hypometabolie (als Ausdruck der Gewebshypoxie) gemeinsam betrachtet werden müssen. Der funktionelle Zusammenhang dieser Reaktionen beim Wirkungsmechanismus des Strahlenschutzes wird erörtert.
