The calorigenic effect of body-own sympathetic compounds and their meaning for the thermoregulation in the homeotherm

Abstract
The sympathetic nervous system and its transmitter substances adrenaline and noradrenaline contribute to the chemical thermoregulation through stimulation of heat production and to the physical thermoregulation through contraction of skin vessels. Both mechanisms are influenced by ganglionic blocking agents and by blockers of the sympatheticα-receptors. These drugs therefore do not permit differentiation between sympathetic influence on heat production and heat dissipation. Propranolol, a new sympatholytic agent acting exclusively on adrenergicβ-receptors, however, does not inhibit the vasoconstricting but the calorigenic action of adrenaline and noradrenaline. Propranolol should therefore be a useful tool for the specific inhibition of the sympathetic influence on heat production. Our experiments showed that this is indeed the case. In awake rats propranolol inhibited completely the increase of oxygen consumption due to artificial lowering of ambient temperature from 30° to 10° C. Heat dissipation in cold (10° C) and hot (37° C) surroundings was not influenced by this agent. In clear contrast to propranolol, chlorpromazine and urethane diminish heat production as well as heat dissipation, both induced by changes of ambient temperature. Confirming results were achieved in anaesthetized rats and normal mice.

Propranolol blockiert ebenfalls die kälteinduzierte (10° C) Steigerung des Sauerstoffverbrauchs (chemische Wärmeregulation), beeinflußt dagegen die Wärmeabgabe in Kälte (10° C) und Hitze (37° C) (physikalische Wärmeregulation) wenig oder gar nicht.

Die durch Propranolol verursachte Thermoregulationsstörung ist also durch eine isolierte Ausschaltung der gegenregulatorischen Wärmebildung gekennzeichnet. Nach Chlorpromazin und Urethan ist dagegen auch die physikalische Wärmeregulation beeinträchtigt.

Die Hemmung der kälteinduzierten Thermogenese durch Propranolol umfaßt nicht nur die extramuskuläre Wärmebildung (non-shivering thermogenesis), sondern die gesamte Thermogenese. Die Ursache der Propranolol-Wirkung auf die muskuläre Wärmebildung bleibt zunächst unklar.

Auch bei Mäusen wird die Steigerung des Stoffwechsels in Kälte (0°C) durch Propranolol verhindert.
